35 Sensors in Medicine 2020 Two peaks indicating fluorescence along the lateral flow assay (LFA). One is caused by the OTR at the miR-150-5p test line and the other is the fluorescence at the control line. DETECTION OF microRNA ON PAPER Integrals of fluorescence scan showing difference between oligonucleotide templated (OTR) and non-templated reactions (NTR): ACKNOWLEDGEMENTS: This project is funded by the March of Dimes. CONCLUSION Fluorescence along LFA using Bench-top Scanner • Preliminary results indicate ability to detect multiple miRNAs on a single strip (multiplexed analysis) whilst also incorporating a control line. • A nM LOD was achieved. • The technology proved sensitive enough for amplification-free detection of endogenous concentrations of microRNAs from clinical samples and for efficiently discriminating between women who delivered at term and those who ended up delivering preterm 3 . The fluorescence increases with increasing concentration of miR-150-5p. A similar fluorescent signal is produced in different conditions (no microRNA, non- complimentary as well as complementary nucleic acids), as the fluorescence is microRNA- independent. miR-150-5p Control line
