Sensors in Medicine 2020 4 Immunoaffinity Monoliths for Extraction of Multiple Preterm Birth Biomarkers from Blood Serum in 3D Printed Microfluidic Devices Haifa M. Almughamsi , Karyna M. Woolley Department of Chemistry and Biochemistry, Brigham Young University, Provo, UT, USA ABSTRACT Preterm birth (PTB) is the leading cause of neonatal deaths. Currently, no biomarker-based methods are readily available to determine the risk of PTB prior to onset of labor. We are creating 3D printed microfluidic devices with immunoaffinity monoliths. These monoliths are modified with antibodies to extract and enrich the PTB biomarkers from human blood serum. We have successfully extracted ferritin from human blood serum 1 . This approach was expanded to extract three additional PTB biomarkers. This work shows important progress toward a rapid diagnostic that is capable of purifying all nine PTB biomarkers from human blood serum. IMMUNOAFFINITY EXTRACTION Monolith formation 1 • Monomer: 24% glycidyl methacrylate (GMA) • Crosslinker: 11% ethylene glycol dimethacrylate • Porogens: 10% cyclohexanol and 55% 1- dodecanol • Photoinitiator: 1% 2,2 dimethoxy-2-phenyl acetophenone Monolith is exposed to UV light for 10 minutes to polymerize.
